Almost every research peptide is sold as “≥99% pure”. The number is real, but it answers a narrower question than most buyers assume. Here is what HPLC purity measures, what it misses, and the other tests you need before you can trust what is in the vial.

What HPLC purity measures

High-performance liquid chromatography (HPLC) pushes a dissolved sample through a column that separates molecules by how strongly they interact with the column material. A detector, usually UV absorbance, records each component as it comes out, giving a chart of peaks called a chromatogram.

Purity is the area of the main peak divided by the total area of all peaks. A result of 99.2% means 99.2% of the UV-absorbing material the method detected came out as the main peak. The other 0.8% is impurities, typically truncated sequences, deletion sequences or by-products from synthesis.

What HPLC purity does not tell you

  • Identity. HPLC shows that one compound dominates the sample, not which compound it is. A vial that is 99% pure of the wrong peptide still reads 99%.
  • Quantity. Purity is a percentage, not a weight. It does not confirm a “10 mg” vial holds 10 mg of peptide. Lyophilized powder also contains counter-ions (such as acetate or TFA) and residual water, so peptide content by weight is lower than the total powder mass.
  • Invisible contaminants. Endotoxins and heavy metals do not show up as UV peaks. They need their own tests.
  • Method quality. A poorly designed method can hide impurities under the main peak. Gradient, column and detection wavelength all affect the number.

The tests that complete the picture

Test Question it answers
HPLC purity What share of the sample is the main compound?
Mass spectrometry (MS) Is the main compound the right molecule? It confirms molecular weight against the expected sequence.
Endotoxin (LAL) Is the sample contaminated with bacterial endotoxin, which can disrupt cell-based assays?
Heavy metals Are metals such as lead, arsenic, cadmium or mercury present above limits?

Purity plus mass spectrometry together tell you the vial contains the right compound in a high proportion. Endotoxin and heavy-metal results tell you it is clean enough for sensitive work.

Purity is also separate from net peptide content, the share of the powder that is peptide rather than counter-ions and water. See TFA vs acetate salts, and for where impurities come from, how research peptides are made.

Is 98% very different from 99%?

For many applications the gap is small. It matters more for sensitive assays, quantitative work and long studies, where impurities can add up or interfere. What matters more than one extra point is whether the number is real: tied to a specific lot, tested by a named accredited laboratory, and shown as a chromatogram rather than just a stated figure.

How to check a purity claim

  1. Find the lot number on the product page or vial.
  2. Open the Certificate of Analysis for that exact lot.
  3. Confirm the lab is named and accredited (look for ISO/IEC 17025).
  4. Check that the chromatogram shows one dominant peak and the reported percentage matches it.
  5. Look for a mass-spec result matching the compound’s expected molecular weight.

Our guide to COA red flags lists the signs that a certificate has been reused or edited, and endotoxin and heavy-metal testing explained covers the two contaminant tests in detail.

Our standard

Every lot we sell is tested by Bioviridian Labs, an ISO/IEC 17025 accredited laboratory in Texas, for HPLC purity, mass-spectrometry identity, endotoxin and heavy metals. The current lot and its purity appear under the Add to Bag button on every product page and in our COA Library.
