Most metabolic research compounds act on the incretin receptors GLP-1 and GIP. Cagrilintide works through a completely different hormone, amylin, and a different receptor family. That is exactly why researchers pair it with GLP-1 compounds. This guide explains what amylin does and where cagrilintide fits.
What amylin is
Amylin is a 37-amino-acid peptide hormone released from pancreatic beta cells together with insulin. In research models it slows gastric emptying, suppresses glucagon after meals and acts on areas of the brain involved in satiety. Native amylin clumps easily and is cleared quickly, which limits its use as a research tool.
What makes cagrilintide different
Cagrilintide is a long-acting amylin analogue. It carries amino-acid changes that reduce clumping and a fatty-acid side chain that binds albumin and greatly extends its activity. It acts on amylin receptors, which are calcitonin receptors paired with receptor-modifying proteins (RAMPs), and on the calcitonin receptor itself. Cagrilintide research profile.
Amylin pathway vs incretin pathway
| Cagrilintide | GLP-1 / GIP compounds | |
|---|---|---|
| Hormone mimicked | Amylin | GLP-1 and/or GIP (incretins) |
| Receptor family | Calcitonin receptor + RAMP (amylin receptors) | Class B GPCRs: GLP-1R, GIPR |
| Examples | Cagrilintide | Semaglutide, tirzepatide, retatrutide |
Because the two pathways use separate receptors, combining them can test whether their effects add up, something you cannot do by stacking two GLP-1 compounds.
The cagrilintide + semaglutide pairing
The best-known research combination pairs cagrilintide with semaglutide: one amylin-pathway compound and one GLP-1 compound. A clean study design runs each alone plus the combination, so the combined effect can be compared against each part. Our blends vs single peptides guide explains that design, and semaglutide vs tirzepatide vs retatrutide covers the incretin side.
Handling and testing
Cagrilintide is a long, acylated peptide supplied lyophilized. Store sealed vials at -20°C away from light and see our storage guide. Every lot is tested by Bioviridian Labs (ISO/IEC 17025, Texas) for HPLC purity, mass-spectrometry identity, endotoxin and heavy metals; see the COA Library.
